Jacksonville Jaguars at San Francisco 49ers Preview, Prediction, Pick for Sunday 9/28/25

Both the Jacksonville Jaguars and the San Francisco 49ers will enter this game with records they are pleased with, but confused about how they achieved them. Jacksonville’s struggle to get Brian Thomas Jr. going has overshadowed their 2-1 record, one featuring a big win over the Texans in Week 3. San Francisco’s slew of injuries — including a season-ending ACL tear for Nick Bosa — has dominated headlines over their 3-0 start. Now they welcome back franchise QB Brock Purdy from his turf toe injury just in time for this showdown.

The 49ers, playing at home, are -3.5 favorites for Sunday’s action, and a Jaguars upset is priced at +160 on DraftKings Sportsbook. The total is set at 46.5 points. Let’s dig into a preview, prediction, and pick for Sunday’s matchup between the 49ers and the Jaguars.

The 49ers have found a way to gut out victories no matter what, winning all three games by one score while managing injuries. They fought through the Seahawks in Week 1 despite losing George Kittle and Jauan Jennings. Quarterback Mac Jones led their Week 2 performance in Purdy’s absence. Then, in a last-minute Week 3 win over the Cardinals, Jones cobbled together a game-winning drive after hurting his PCL. The one-legged backup put on a spectacular display of timely clock management to give the Niners the lead in the NFC West.

They’ll have their match in the Jaguars. San Francisco has the second-best defense by PPG at 16.3, but Jacksonville’s not far behind them at 17.0. Both offenses will need to pick it up if they hope to win.

Each team has established different balances. Jacksonville is fifth in rushing yards per game (141.7) and fourth in rushing yards allowed (82.7). The 49ers are first in passing yards per game (271.3) and fifth in passing yards allowed (162). Will they continue on these trends, or will someone be able to break the mold?

Jaguars at 49ers pick, best bet

It’s hard for teams to break loose on the ground against San Francisco. Even though their defensive front took a hit in Bosa’s absence, all-world linebacker Fred Warner is waiting around to clean things up. The lack of Kittle negates Jacksonville’s advantage of having excellent coverage linebackers. Purdy, healthy and ready to go, will lock on to breakout receiver Ricky Pearsall and a chain-moving slot man in Jennings.

Jacksonville’s inability to contain San Francisco’s pass game will be their undoing. Count on the home favorites to deliver.
